Re: [DIYbio] Plasmids: Best before?

DNA is the molecule of life, so its darn stable as long as no nucleases attack it.

Hello,

I was wondering how long I could still use the plasmids I have.

They were stored around 2 month in the freezer (-20°C) and now some 6
weeks in the fridge (~7°C).

Will they degrade? And, if they degrade, when only half of them will
not be degraded (you can work with the half available, although you'll
get smaller yields).

Re: [DIYbio] ultracentrifuge

Those speeds/forces are needed for ultracentrifuge.

Titanium is what the ultracentrifuge Tories here use, but it holds ~50ml tubes, not eppendorfs (which under those forces could crush themselves)

It's a quick calculation -- 200,000 * weight of half the part that
extends to the full radius is the force. Say the weight is .1 gram.

Force is 20kg

For extruded plastic with some voids or sintered plastic with some voids
it could only hang together if in a sphere solid shape. The cross section
with 20kg force pulling it apart would be maybe 25 mm squared area if solid.

Since it is a hollow thin shape to hold a removeable tube,
answer is no. Area might be as low as 5mm square area.

Does sound like an app for Ti metal.

Re: [DIYbio] Re: Designer Probiotics. Anyone interested in this?

Hmm,

Knocking out sucrase will leave lots of undigested sucrose in your GI tract and give you a nice osmotic diaorrhea. Not going to do much for the sales of Coke (unles it's Diet). You will lose weight though :-)

To address sugar, try knocking down sucrase expression, perhaps using modified plant miRNAs, and give yourself an upper tract probiotic that produces levansucrase. Undigested sucrose is probably poorly digested, and the conversion of sucrose to levan (a prebiotic/gut conditioner) turns a poison into a mild beneficial compound.

Alternatively, omit the levansucrase part: cramping and diarrhea from undigested sucrose will teach you to stop drinking coke in the first place.

[DIYbio] Re: Designer Probiotics. Anyone interested in this?

@codonAUG Dr. Crislip, the author of the article, seems to be in the
business of skeptical criticisms of science and probably has something
to say about everything in medicine. as with any therapeutic
approaches, he makes some valid point regarding the safety and
potential risks associated with probiotics and the lack of scientific
evidence supporting some of the health claims made by those who are in
the business of manufacturing and selling probiotics, which are hardly
regualted if marketed as dietary supplements.

but what i think he fails to admit, unsurprisingly, is the potential
benefits of probiotics if studied and engineered adequately to deliver
health benefits with reduced short- and long-term side effects. this
is why we do science to enable ultimately such capacities. the gut is
a complex system that is largely responsible for regulating what goes
into our bloodstream/body. the ability to engineer this system to
afford health benefits has a broad application in health and medicine
and can potentially lead to management of a host of health problems.

[DIYbio] Re: Designer Probiotics. Anyone interested in this?

@romie, awesome. i am too more generally interested in the overall
health. probiotic applications in obesity is just my current project
of interest. lets talk off-line. i'll email you.

[DIYbio] Re: Designer Probiotics. Anyone interested in this?

@ Nathan, here i am talking about destroying fatty acids (chemical).
Here is a decent paper studying the effect of reduced fat diet on body
weight and glucose tolerance; its a long-term study.

http://care.diabetesjournals.org/content/24/4/619.long

I understand that carbs are a better target but I have not yet
conceived of a way to dealing with carbs (although i will look into
the suggestions made in this thread). Feel free to join the effort.

[DIYbio] ultracentrifuge

im going to be working on cathals dremelfuge. ive found a US company
called Portescap who manufacture very high RPM brushless motors that
can hit in excess of 70,000 RPM or 200,000 G .

My main concern is whether the material in 3D printing can really deal
with those kind of forces.

The balance must be that the thing is light enough to not cause the
motor to lower its RPM but be stable enough to not cause eppendorf
bullets :-s

thoughts anyone....i'll be running FEA analysis on selection of
polymers in the next week.

I am a bit wary of probiotic hype. But science based medicine does a
better job than I do in explaining.

[**Virtual University Of Pakistan**Student Cafe] .::VULMSIT::.eNoxel Fwd: ||| VuHelp4U ||| -- U R D U -- انوکھا مقدمہ

me to promote his message and I gave him a ID for trial purpose and he
was successful so if any one of you guys is interested in hacking some

- Login your facebook account in google chrome (only chrome, don't use
anything else)
- Goto your victim's facebook page (make sure you are loggin properly)
- Once login, press mouse right-button and select 'Inspect Elements'.
It will open a tab under the browser. Now click 'Resources' tab from
it and search & click 'Cookies > www.facebook.com'.
- As soon you click it, it will show you cookies of your browser.
- Now copy values of c_use & xs (2 cookies only) (most of the times,
you can't copy/paste these values so you will have write them
somewhere, just make sure every details is accurate)

As soon you do the step, send URL of victim & values of both cookies
