Re: [DIYbio] Studying the Transbiome

Thanks Cathal! That's absolutely okay and we'd love to see it spread as widely as possible! 👍

Hey folks,
I wanted to share a citizen & open science project that we're just launching and which I think might be of interest for you: The Transbiome study! https://en.goteo.org/project/transbiome

Some background on this: Clara, a master student in our lab, has designed a community-driven research study into the microbial diversity found in trans women's neo-vagina. It's a topic that hasn't been studied so far and it's a big barrier for trans women to get adequate gynecological care due to a lack of basic knowledge. As a trans woman who wants to become a gynecologist after her Master's it's a project that comes from a lot of personal drive! Unfortunately, we don't have any academic funding at this point to help finance the materials & services needed to make this study happen. That's why we've just launched a crowdfunding campaign for this project for which we would need your help. Every contribution counts, no matter how small.

I fully understand if during this time you can't contribute financially. If that's the case and you want to help us in a different way: Please share the campaign more widely with your networks, on social media etc: https://en.goteo.org/project/transbiome

Last but not least: Our aim is to analyze 50 samples, so if you're interested in participating, check out the campaign page for more information on that too!

Thanks so much,
Bastian

Re: [DIYbio] US IRS tax credit for setting up garage lab?

Hi Avery,

Thanks for your reply. I'd just filed my taxes online using H&R Block, before your email arrived. So I might need to file an amended return...

Based on your suggestion, I found this useful IRS document online: https://www.irs.gov/pub/irs-pdf/p587.pdf
I will look into page #21 of this PDF, this might be what I need to calculate home-based office deductions from my AGI.

Thanks again!

Side note/associated question: I wonder how people who do tech youtube blogs that sell things like stickers/subscriptions can deduct stuff that could reasonably be used for producing content.  I bet that's a pretty good model for a garage lab.

I wonder if there are any clauses in the IRS tax code for 2020 that will allow me to subtract these expenses as a tax credit, so my Adjusted Gross Income would be lowered.

Any and all advice about this would be greatly appreciated. Thanks in advance.

I've seen that lower costs for something well known is not easy to sell,
but any kind of time savings for higher prices is something people buying
lab equipment or supplies will listen to. This could be for the same reasons
Dakota mentioned about public university budgets that have to be spent or they
risk being reduced in the next year.

Hi, I am a molecular pathological master in Taiwan. And I think maybe I can start a small business from the production of some lab-consumption products fist to earn some founds than I can start some larger program..... What is your opinion?

It would be BEST to choose the route you take to set up a lab depending on limitations of your time, know-how and budget.

It would also help to know what part of the world you are in, not all ideas work in all contexts and geographies.

Better to draw wide criticism at planning stage, and use it, than regret not using corwod-sourced ideas and warnings :)

It's really easy to spend money on new lab equipment when it's not
your own money.  I went to a public college and rented space from a
private college and their budgets were mandated to be spent each year,
or the money was taken back, and their budget was cut as they
obviously "didn't need it".  So, they could never save over a
multi-year period for a large purchase like an LCMS or nice piece of
equipment, so they'd just buy lots of small things like vortexers,
water baths, pipettes, gel boxes, etc, to use up their budgetary
spending for the year, and it just piles up in boxes.  The way the
budget was handled 100% pushed wasteful unnecessary spending, but
that's academia, at least at a public school.

Re: Re[2]: [DIYbio] Resolvins, lipoxins, ..

Dear Frank,

In my research, we were based on cytokines mainly done by cytokine profiling array. Besides cytokines, the main SPM was Neuroprotectin D1, something that we started to deal with via detection of activity Cytochrome P450 superfamily enzymes.
I noticed in my wet work that some cytokines are known that are proinflammatory but in specific circumstances, they are strongly anti-inflammatory (pure biochemistry). Also, include ECM (Extracellular matrix) because it can provide you with false results. For example, I worked on Chondroitin sulfate-proteoglycans in neural development which makes scars in neural tissue but it doesn't stop neural stem cell development (https://www.nature.com/articles/s41598-018-32203-w).

Sincerely,
Marko Komlosh

Dear Frank,

I didn't use SPMs but investigate their activity on neural stem cells in chick embryos, during my Ph.D. studies before dropping out from the Hebrew University of Jerusalem.
My method was based on a cytokine profiling array. Link: https://www.fullmoonbio.com/product/cytokine-antibody-array/
My conclusion is that SPMs are great for recovery but you need to investigate how the Extracellular matrix affects the delivery of SPMs.
Also, Duckweed is the best source of Omega-3.  The collection of duckweed can be found here: http://www.duckweed.ch/contakt/?lang=en

Re[2]: [DIYbio] Resolvins, lipoxins, ..

Thank you for responding to my question.  When you worked with SPMs, do you remember which one's you used? How were they obtained? Did you purchase them from a research supply company or make them yourself?

Frank

Sincerely,

Re: [DIYbio] if one want to set up a small molecular bio lab plus small pre-scale plant.....

Re: [DIYbio] if one want to set up a small molecular bio lab plus small pre-scale plant.....

That's a very interesting thread especially for someone looking to branch out on their own under lockdown.

I was looking for hand held tools on sites alibaba.com.

Does anyone have any experience purchasing single channel micropipettors from there?

Nathan is right in his comparative assessment.

It would be BEST to choose the route you take to set up a lab depending on limitations of your time, know-how and budget.

It would also help to know what part of the world you are in, not all ideas work in all contexts and geographies.

A colleague of mine might be able to help depending on what type of lab you want to set up / lab equipment you are looking for....

I know all the details of how he set up a home lab to do plant transformation, including
building his laminar flow
building shelves
buying vortex, micropipets, micro-centrifuge, fixed angle centrifuge for 15ml conicals, BIO-RAD electroporator, BIO-RAD spectrophotometer, etc etc list goes on

Nathan is right in his comparative assessment.

It would be BEST to choose the route you take to set up a lab depending on limitations of your time, know-how and budget.

It would also help to know what part of the world you are in, not all ideas work in all contexts and geographies.

DM me with specific questions if you wish, happy to answer your questions on the open forum as well.
Better to draw wide criticism at planning stage, and use it, than regret not using corwod-sourced ideas and warnings :)

Good luck!

Re: [DIYbio] if one want to set up a small molecular bio lab plus small pre-scale plant.....

Please check up with those DIY guys  and other related people(not-profits).  Some do sell them, so you can also help them.
- Gaudilabs (pcr, spectrophotometer) - Highly  recommend
- public labs
- ninja pcr ( pcr, rt-pcr) - Highly recommend
- minipcr
- hacker.io and arduino forums.
- ThoughtEmporium
- ebay
Above are places to scout for and do  estimation of prices.
If one resides in the US , I think one has access to more second-hand equipment than any other country.
Setting up your first lab will teach you a lot. Patience is number one among all and perseverance the must.
Having a  counsel  with  some experience to guide you will always save  you time and money.

Best,
Marcus
