The standard hello world is to do a bacterial transformation. Simply put, you get some bacteria and add a new gene to them. Procedure can be found here: http://tequals0.wordpress.com/diybio-protocols-and-information/
I am also giving away most of the "expensive" (relative to one person vs quantity needed) reagents to do this. Mail me off list for details.
