Hey
-- A little less than a month ago I posted on -20 storage. I quote from it:
"However, nearly 1/3 to 1/2 of these cells no longer held the RFP plasmid even though I plated on amp"
I had originally thought these to be satellite colonies but as the plate sat on my bench I noticed that there was a white colony far away from any red colonies. So I streaked out 4 white colonies and 4 red colonies onto 2 plates (Picture is attached). Turns out they were still resistant to Amp
This might have been since RFP put a metabolic burden on the cell and so removing it or turning the gene off made the cells grow faster. I am going to work on identifications of the mutations this week.
