Dear DIY community,

Our garage lab is now switching from plastic disposable peri dishes to glass non-disposable petri dishes for microbiology work. The glass petri dishes are yet to arrive, but never having used the latter, I have some questions I need your help with, please:

My dad is recovering very well in hospital from COVID, and told me to reach out to DIY and other researchers to learn answers I do not know, instead of feeding them to me, so I can learn the logic of each protocol and step in research. Therefore, I am emailing you from his account. Though I am only in the 6th grade, I have some lab experience, but feel free to explain in elementary language and as much detail as you wish.

In your response, please do share any reading material, or video links, as well as product links for where I can purchase any missing lab equipment for my purpose. Thanks, in advance!

Question 1

In what does one put the petri dishes into while autoclaving (in our simple yet effective steam pressure cooker)? Is there a cylindrical metal jacket that folks have used in the past? Just a vague memory of seeing this in a lab somewhere, but not sure and cannot remember clearly. Or find anything online.

Question 2

During autoclaving, should I wrap the plates (lid + bottom) individually in Al foil?

Or even separately wrap the lid and the bottom?

Question 3

And finally, should I 

only steam autoclave ?

only dry autoclave?

first steam and then dry autoclave?

We've been getting mold in our plant tissue culture medium containing sucrose after ~ 2 weeks despite use of a certified sterile laminar flow and other sterile techniques, so I am inclined to do steam + dry in the same cycle...
