my biggest issue with microscopy at the moment is being able to take nice photos, especially at oil immersion levels. I can't remember if my oil immersion lens if 400x or 1000x, and that's because I never use it...because I never have nice oil, and the few times I've tried cooking oil, my "in focus sphere" is so small...I can barely make out anything.
Right now I use my camera which I stick inside a toilet paper roll to serve in place of some focal length modifier...and it works OK but sometimes the field of view is narrowed...or the light levels are low.
Being able to throw a sample in a scope...click a button, and take a picture that goes directly to the computer would be awesome.
Right now I have to fiddle with the sample, then the toilet paper roll and the focus on my camera, then the light levels, then I have to connect my camera to my computer, upload, find the pictures, etc
So mainly, I'd say the things I find most important are taking decent pictures that go directly to computer/internet easily, and the ability, as Avery said, of having nice 400x +++ pictures to be able to see bacteria.
