Also make sure that your strain has not developed amp resistance due to exposure/mutation of plasmid.

I did several tries with the ampicilling in different concentrations and can conclude:
The amp does not work because there is a lawn of bacteria on each plate!!!!
I'll try again with 500mg of amp-powder. Maybe it's just far too weak. Hopefully.
