For a traditional molecular biology lab your looking at a cost roughly $30,000 new. However, that's unrealistic for the DIYbio community. How much are you looking to spend for your DIYbio lab?

[DIYbio] Plant genetic transformation visible marker help

Hi folks!

Would any of you happen to have plasmid DNA constructs that work as visible markers of plant genetic transformation, without the need for complicated equipment for screening fluorescent (GFP, DsRed, mCherry etc) protein screening OR without the need for destructive assay like the traditional GUS staining protocol ?

Something like 
RNAi against phytogene desaturase - bleached, white leaves
and / or
overexpression of anthocyanin - purple colored plant tissues
would be idea.

Please let me know. Any and all ideas welcome! Thanks, in advance.

So back in the spring, I took a survey of people into DIYbio, and I chose 2 participants at random to receive $50 Amazon gift cards. One of them was successfully received, but the other still just says "Sent" (as per the screenshot - not completely current, but that's still how it looks on my Amazon account). [DIYbio] Help needed with shRNA interference....

Dear DIYbio,
        I've reverse engineered a short hairpin RNA sequence that produces an interfering RNA that I want to express in human cells. 



This RNAi targets myotilin, which when misfolded causes a very rare form of muscular dystrophy. Eliminating the misfolded myotilin largely reverses the disease symptoms in both mice and rhesus monkeys. No side effects from the loss of myotilin were observed.
         I now have no idea how to proceed with getting AAV to express the above. The design, and manipulation of plasmids is beyond me, so I'm happy to ask a plasmid factory to do this for me, it's just that I don't know what to ask for......
Any help would be appreciated.....
