Trying to root my Christmas tree, thinking about LED lighting for plants

I welded a tree stand and put the tree up about 9 or 10 days ago,
cutting about 2cm from the bottom and a few of the lowest limbs as
well. A steel bowl was placed under the tree trunk and a small chunk
of wood was used as a standoff. The standoff was smaller than the
diameter of the trunk, thinking that I would prefer to stress the
heartwood rather than the vasculature. A single bolt was driven into
the trunk 1-2cm for support, and guy-wires (Romex) were attached ~120
degrees relative to the bolt. (My welding equipment isn't proper and I
slacked off toward the end because things were taking so long)

The first night I soaked the tree in water with about half the
recommended strength of rooting solution ("" fertilizer with IBA (an
auxin) in it).

The next day I added soil to the brim of the bowl, and filled to a
muddy consistency with more water+rooting solution. I checked the
water level 2-3 days later and topped it off, then left home for 4
days, and topped it up as soon as I got home.

Since we put the tree up its had a strand of white LED Christmas
lights on it, and I'll try to get some more tomorrow. Now I'm thinking
about the best way to do this... i.e. what wavelength or mix of
wavelengths is best for glucose production?

dealextreme has some LED string lights (100, 150, 192, 300, 600 LEDs
per string) with different colors, some list the wattage (which
reviews sometimes say are incorrect) or mcd output or wavelength or
color temp...

I also found this paper that says 720Hz pulsed LEDs increased growth
in potato plantlets over continuous-nonfluctuating (C-NF) mode.

Any suggestions? Maybe get a blue and a red strand of the SMD lights
from dx and build a controller to pulse each at 720Hz??

##hplusroadmap is mostly amateur biologists, believe it or not.


There's about 50/day active on average and a few years of history of not being dead. There's also #biopunk somewhere, but it's mostly inactive like #diybio.

Re: Get a plasmide - buy? share?

An alkaline miniprep is basically an acid/base (A/B) extraction where the point is to "defat" the cell solution using detergent to act as the nonpolar component... it diverges from a normal A/B extraction because you don't separate nonpolar from polar before neutralizing... thanks to the size of chromosomes/nucleosomes they get caught up with the nonpolar stuff and pellet out.

Lysozyme isn't needed, but helps. Sugar in buffer is probably just for osmolarity so cells don't burst before being alkalinized, because the alkalinity deactivates nucleases. Triton-X is a line of fancy and well documented detergents, so protocols have been refined to make sure its all cleaned from the plasmid solution... other detergents will work, but the protocol may be lossy without lots of experimental optimization. EDTA is also there to stop nuclease activity but again not absolutely required... unless you plan on storing the plasmid for a while or at room temperature.

Re: IRC Channel

The problem with IRC is that only a small subset of the user-base uses
it in the first place. And we need the place to be populated most of
the time so it doesn't feel so useless. I use irc once in a while when
I got some question about programming, ubuntu, or those few times I
feel like playing "magic the gathering". The mailing list seems to get
more faster replies.

I'm not sure I went to the right place but the channel you made was

Greetings,

I am rather new here, though I have been lurking for a while. I
thought it would be interesting to get to know some of the people in
the DIYbio community, and in another group I belong to, we have an IRC
channel. I think it allows discourse in less topic-relevant subjects.

Looking through old posts, I was curious if there were any active IRC
channels associated with DIYbio. My searches on the google group
yielded #diybio on irc.freenode.net. However, this appears to now be
occupied by a small number of transhumanists, where I am looking for a
group more based on citizen science/diybio/amateur biology.

I grabbed the channel #diybio on irc.darkmyst.org, if anyone cares to
join. If there already is an active channel, can you direct me to it?
